| Abstract & Background | Abstract In the research of DNA computer, immobilization of DNA molecule is an essential technology at first. Actually, immobilization of DNA molecule is not only a basic tool of biological computation, but also the foundation of the whole Genetic Engineering and bio-chip, even some disease treatment. In this paper, immobilization technology of DNA molecule, including its basic principle and factual methods is discussed in detail. The architecture of bio-molecular immobilization system is presented. The authors mainly introduce its three subsystems¡ªsubstrate system, fixer system and molecular marker system. It is often used to detect and analyze DNA molecule, purification or gene sequencing. In the process of DNA computing, DNA molecules are often needed to be immobilized on the surface of tube, gel, glass or other substrates in order to be detected accurately. With the development of bio-sensor recently, more and more DNA computing methods based on DNA sensor are presented, and immobilization of DNA molecule is also an essential technology of DNA sensor. In addition, immobilization of DNA molecule is not only a basic tool of biological computation, but also the foundation of the whole Genetic Engineering and bio-chip, even some disease treatment. This research is supported by National Natural Science Foundation of China (60910002, 60974112, 60971085, 30970969), the National High Technology Research and Development Program(863 Program) (2009AA012413), Ph.D. Programs Foundation of Ministry of Education of China (20070001020), and Postdoctoral Science Foundation of China (No.20080440257).The projects mainly focus on DNA computer models for processing graphical messages, including encoding DNA sequences, synthesizing DNA molecules, setting up the model, detecting solutions, etc. The research group has been working on many aspects of DNA computing since 1996. They have published a monograph and more than 100 papers on DNA computing. In this paper, immobilization technology of DNA molecule and its application are introduced in detail, this technology is an important factor which affects the efficiency and accuracy of DNA computing. |
